Week 2: September 19, 2022 - September 23, 2022

September 20, 2022  Set up for Lower Kanamycin Concentration Testing   Since D.aquaticus only showed growth with Kanamycin, we set up to test five lower concentrations of Kanamyacin. First we made 250mL of TGY by combining tryptone, yeast extract, dextrose, and distilled water. It went in the autoclave overnight and the next day, the concentrations were prepared. There was two samples of each concentration in the end. Concentration one contained 3.125 μL /mL of antibiotic. Concentration two contained 2.733 μL /mL of antibiotic. Concentration three contained 2.34 μL/mL of antibiotic. Concentration four contained 1.95 μL/mL of antibiotic. The last concentration, number five, contained 1.56 μL/mL of antibiotic.      My first week in the lab, I joined a group that is testing to see if  Deinococcus aquaticus has a gene that is resistant to one of three antibiotics. The three antibiotics they are testing are tetracycline, kanamycin, and chloramphenicol. More specifically, they are testing to see at which concentration of antibiotic D.aquaticus can still continue to grow.  The Three Antibiotics Tetracycline is used to treat infections caused by bacteria. It works by preventing the growth and spread of bacteria. This antibiotic is typically prescribed to treat pneumonia and other respiratory tract infections, certain infections of the eye, skin, lymphatic, intestinal, genital, and urinary systems. Tetracycline can be prescribed, along with other medications, to treat acne or to treat patients who can’t be treated with penicillin in order to treat certain types of food poisoning.
