Week 7 - October 28, 2022

 Tuesday October 25, 2022 

Making Media:

On Tuesday, we were asked to make media for the microbiology department. There were five different types of media that my partners and I were asked to make. On this day, we began by making the first three. We first filled test tube racks with test tubes. Each test tube had a Durham tube placed inside with the open side down.


Phenol Red Manitol Broth 2L:

  • 21 g per liter of Di Water 

  • Stir to dissolve the

  • Use a beaker to dispense/pour media into tubes

  • Cover the Durham tube with media 

    • This is approximately 7 ml 

  • Place cap on loosely 

  • Label the test tube rack clearly with tape

    • I used one on each of the longer sides and one piece of tape across the top of the tubes

Phenol Red Sucrose 2L: 

  • 21 g per liter 

  • Stir to dissolve the

  • Use a beaker to dispense/pour media into tubes

  • Cover the Durham tube with media 

    • This is approximately 7 ml 

  • Place cap on loosely 

  • Label the test tube rack clearly with tape

    • I used one on each of the longer sides and one piece of tape across the top of the tubes

  • 21 g per liter of Di Water 

  • Stir to dissolve the

  • Use a beaker to dispense/pour media into tubes

  • Cover the Durham tube with media 

    • This is approximately 7 ml 

  • Place cap on loosely 

  • Label the test tube rack clearly with tape

    • I used one on each of the longer sides and one piece of tape across the top of the tubes



We did not get to finish pouring all of the media into the tubes so we set them aside and they were finished on Wednesday by other group members. The racks that were finished were set aside to be taken to where they needed to be. 



Thursday October 27, 2022 

Making Media Continued 


Phenol Red Glucose 2L: 

  • 15 g Phenol Red Broth Base 

  • 5 g Glucose 

  • 1,000 ml Di Water 

  • Stir to dissolve the

  • Use a beaker to dispense/pour media into tubes

  • Cover the Durham tube with media 

    • This is approximately 7 ml 

  • Place cap on loosely 

  • Label the test tube rack clearly with tape

    • I used one on each of the longer sides and one piece of tape across the top of the tubes

 MRVP 500 ml: 

  • 8.5 g per liter 

  • Stir to dissolve the

  • Use pipette to dispense 2 ml into test tubes and cover them with metal caps

    • These tubes did not require any Durham tubes  

  • Label the test tube rack clearly with tape

    • I used one on each of the longer sides and one piece of tape across the top of the tubes


After making the media for the microbiology department, I went on to make media for our D. aquaticus project. I made 250 ml of TGY by mixing together 0.75 gram of Tryptone, 0.75 grams of Yeast Extract, and 0.25 grams of Dextrose. It was then put into the autoclave.
